Part III, Mathematics, natural sciences and technology"}]}]},"item_16_description_4":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"緑色植物の先祖型と類似していると考えられている原生生物,灰色藻Cyanophora paradoxaの遺伝子\nを生物進化の視点から解析することは,最初に起こった葉緑体前駆細胞の共生から現在の陸上緑色植物に至\nるまでの進化の過程を探る手がかりになると考えられる。本研究では,原核生物から真核生物が進化する際\nに創生された形質であるステロール生合成遣伝子,中でもsqualene epoxidase遺伝子であるSEに注目した。\nCyanophora paradoxaのSE構造を,進化的な位置付けから確認するために,National Center for\nBiotechnology InformationのESTデータベースからCyanophora paradoxaのSEと考えられる塩基配\n列をBLAST検索によって見出した。推定アミノ酸配列のアラインメント,二次構造や疎水性度の比較を他\nの生物のSE配列との間で行い,Cyanophora paradoxaのSEである可能性及び進化系統樹での位置を検\n討した。また,Cyanophora paradoxaから不ケン化物を抽出し,紫外部での吸収スペクトルを測定するとともに,薄層クロマトグラフィーも行い,ジギトニン反応物としてステロイド骨格構造を予測した。その結\n果,原生生物Cyanophora paradoxaは,B環に不飽和結合を2つ持つ菌類のエルゴステロール型ではな\nく,高等生物と共通した,B環に不飽和結合1つのステロールを生合成しており,また進化系統樹でも高等\n植物に非常に近い類似‘性を有している一方,SE酵素の疎水性度は菌類によく類似していた。 \n------------------------------------------------------------------------------------------\nIn this study, for the purpose of elucidating the evolution process of plant and chloroplast, we\nfocused on Cyanophora paradoxa sterol biosynthesis genes, especially squalene epoxidase gene,\nSE, which produces a rate-limiting enzyme in sterol biosynthesis pathway. SE is located in nucleus\nDNA, and, in a sterol biosynthetic process, protein product of SE functions to prime formation\nof steroid structure from squalene. We found out SB-like base sequence from protist EST database\nof National Center for Biotechnology Information through BLAST program. We compared the\namino acid sequence, hydrophobicity, and the second structure of SE protein from other species\nwith that of Cyanophora paradoxa. In addition, we extracted sterol as unsaponifiable material as\nwel as digitonin-reactive steroid from the protist followed by the measurement of the absorption\nspectra and the profile of thin layer chromatography.
